چکیده

Heme proteins, such as hemoglobins and cytochromes, play important roles in various biological processes. Here we employ the two-photon excited photothermal effect as a contrast mechanism to map heme proteins distribution. Particularly, both a thermal lens scheme and a high-frequency modulation are utilized to enhance the signal-to-noise ratio. We demonstrate label-free imaging of individual red blood cells, subcellular distribution of cytochromes in live mammalian cells, and the microvascular networks in mouse ear tissue and in a zebrafish gill. © 2010 American Institute of Physics. doi:10.1063/1.3308485

Two-photon excited hemoglobin fluorescence provides contrast mechanism for label-free imaging of microvasculature in vivo.

Direct visualization of microvasculature provides significant insights in microcirculation and critically impacts the diagnosis and treatment of microcirculatory diseases. Recently, we discovered that the high-energy Soret fluorescence of hemoglobin peaked at 438 nm with an extremely short lifetime becomes strongly visible under two-photon excitation. Based on the distinct spectral and temporal...

Time-resolved detection enables standard two-photon fluorescence microscopy for in vivo label-free imaging of microvasculature in tissue.

We conducted a systematic study on two-photon excited fluorescence (TPEF) of hemoglobin using the near transform-limited and Gaussian-shaped femtosecond pulse sources. We found that the two-photon action cross section of hemoglobin drops over 2 orders of magnitude in the wavelength range from 550 to 800 nm, while the spectral and temporal characteristics of hemoglobin TPEF are insensitive to th...

Depth-resolved mid-infrared photothermal imaging of living cells and organisms with submicrometer spatial resolution

Chemical contrast has long been sought for label-free visualization of biomolecules and materials in complex living systems. Although infrared spectroscopic imaging has come a long way in this direction, it is thus far only applicable to dried tissues because of the strong infrared absorption by water. It also suffers from low spatial resolution due to long wavelengths and lacks optical section...

Two-photon excited hemoglobin fluorescence

We discovered that hemoglobin emits high energy Soret fluorescence when two-photon excited by the visible femtosecond light sources. The unique spectral and temporal characteristics of hemoglobin fluorescence were measured by using a time-resolved spectroscopic detection system. The high energy Soret fluorescence of hemoglobin shows the spectral peak at 438 nm with extremely short lifetime. Thi...

In vivo micro-vascular imaging and flow cytometry in zebrafish using two-photon excited endogenous fluorescence.

Zebrafish has rapidly evolved as a powerful vertebrate model organism for studying human diseases. Here we first demonstrate a new label-free approach for in vivo imaging of microvasculature, based on the recent discovery and detailed characterization of the two-photon excited endogenous fluorescence in the blood plasma of zebrafish. In particular, three-dimensional reconstruction of the microv...
